研究領域

黃人則老師研究團隊目前的主要目標有 (一) 闡明蛋白質聚集體在神經退化性疾病之致病機轉中扮演的角色,並發展新穎胜肽藥物治療方針;以及 (二) 研究蛋白質共轉譯過程中,其摺疊、導向及運輸可能機制。

在許多神經退化性疾病的細胞中,都發現有不正常蛋白質聚集體的存在,它們共同的特質包含纖維狀型態、富含 beta 摺版的二級結構,但這些蛋白質聚集體的分子形成機制、在細胞層次上如何影響疾病進程,仍待進一步探究。TDP-43 蛋白質被認為是肌萎縮性側索硬化症 (ALS;漸凍人) 和腦前額顳葉退化症 (FTLD) 的致病因子,黃老師團隊過去的研究顯示含 TDP-43 蛋白質中 C 端特定片段序列之胜肽,能夠透過晶種效應加速 TDP-43 蛋白質聚集化並形成纖維狀聚集體。

目前,黃老師團隊透過合成具「光控制性質」的胜肽,配合即時偵測細胞影像之技術,發展一套化學生物學工具,以探討細胞中 TDP-43 蛋白質及其他神經退化性疾病相關致病蛋白 (如杭丁頓氏舞蹈症中之 Huntingtin) 的詳細特性與行為。光控制胜肽在適當波長的光照射之下將斷裂,並開始自組裝聚集化,或誘導其他細胞內蛋白質聚集化。透過準確調控照光之時間點及空間範圍,我們能夠更仔細研究這些蛋白質聚集體以及它們在相關疾病中扮演的角色。目前具體研究方向包含:

    1. 光控制胜肽之設計與合成
    2. 設計新穎螢光染劑
    3. 利用神經細胞株/人類誘導幹細胞研究特定蛋白質在相關疾病中的角色
    4. 結合奈米科學,將胜肽修飾於奈米粒子表面,研究並開發高效率胜肽藥物輸送方式以及生物感測器。

另一方面,疾病中蛋白質聚集體的形成與蛋白質錯誤摺疊亦有關連。目前已知,活體細胞中蛋白質的合成及其正確摺疊受到各種不同類型的伴隨蛋白 (chaperon) 所調控,此過程的錯誤將會導致蛋白質不正常堆疊而進而疾病。因此,黃老師研究團隊也針對蛋白質轉譯過程中,核醣體和伴隨蛋白之間的交互作用關係進行研究。

實驗室成功建立之化學生物學平台,能夠製備螢光標記胺基酸-tRNA 並將之用於無細胞系統進行蛋白質轉譯,表達在任意特定位置具有螢光標記之目標蛋白質。利用該技術,目前研究已經指出伴隨蛋白 Trigger Factor 與 DnaK 會對新生蛋白質-核糖體複合物的動態結構有不同之影響,也正進一步探討兩者對於蛋白質摺疊機制之作用。這些各式伴隨蛋白之研究,將啟發針對與蛋白質錯誤摺疊相關疾病之新式藥物設計。
